Methods&nbsp; Eighty SD rats were selected, among which, 10 were fed with normal diet and taken as blank control group (“blank group” for short), the other 70 rats were fed with high fat and high glucose food and injected with small dosage of STZ intraperitoneally, and 30 models were established successfully. The models were randomly divided into acupuncture treatment group (“acupuncture group” for short), model control group (“model group” for short) and western medicine group (metformin) with 10 rats in each group. After treatment for 4 weeks, RBG, and the expression levels of CHOP, IRE 1, Bax, Bcl-2 protein and mRNA in pancreatic tissue of rats were determined. Result&nbsp; Compared with the conditions before treatment, RBG reduced obviously in the acupuncture group and western medicine group (P&lt;0.05), and there was no statistical difference in RBG in the model group and blank group (P&gt;0.05); the expression levels of CHOP, IRE 1 and Bax proteins in pancreatic tissue of rats in the model group were significantly higher than those in the blank group (P&lt;0.05), the expression results in the acupuncture group and western medicine group were significantly lower than those in model group (P&lt;0.05); the expression levels of CHOP, IRE 1 and Bax mRNAs in pancreatic tissue of rats in the model group were significantly higher than those in the blank group (P&lt;0.01), the expression results in the acupuncture group and western medicine group were significantly lower than those in the model group (P&lt;0.01); the expression level of Bcl-2 protein in the model group was significantly lower than that in the blank group (P&lt;0.05), the expression results in the acupuncture group and western medicine group were significantly higher than that in the model group (P&lt;0.01); there was no statistical difference between the acupuncture group and western medicine group (P&gt;0.05); the expression level of Bcl-2 mRNA in the model group was significantly lower than that in the blank group (P&lt;0.01), the expression results in the acupuncture group and western medicine group were significantly higher than that in the model group (P&lt;0.01); there was no statistical difference between the acupuncture group and western medicine group (P&gt;0.05). Conclusion&nbsp; There was endoplasmic reticulum stress in the pancreatic tissue of rats with STZ-induced diabetes mellitus, and IRE 1-CHOP signal pathway was the main pathway for inhibiting the endoplasmic reticulum stress in the pancreatic tissue. Acupuncture can improve the endoplasmic reticulum stress in pancreatic tissue, inhibit apoptosis genes, and protect the pancreatic tissue through IRE 1-CHOP signal pathway.</p><p><br/></p><p><strong>KEY WORDS</strong>: acupuncture; apoptosis; diabetes mellitus; inositol requiring protein-1</p><p><br/></p><p><strong>ABSTRACT IN CHINESE</strong></p><p style="text-align: justify;">[摘 要] 目的:探讨针刺对STZ糖尿病大鼠胰腺组织内质网应激IRE1-CHOP信号通路及相关凋亡基因的影响。方法:SD大鼠80只,选取10只用普通饲料喂养作为空白对照组,其余70只用高脂高糖饲料喂养和腹腔注射小剂量STZ制作糖尿病模型。造模成功30只,随机分为针刺治疗组模型对照组、和西药组(二甲双胍),每组10只。治疗4周,测大鼠的RBG,胰腺组织CHOP、IRE1、Bax、Bcl-2蛋白和mRNA表达。结果:与治疗前比较,针刺组和西药组RBG均明显降低(P<0.05),模型组和空白组RBG变化的差异无统计学意义(P&gt;0.05);模型组胰腺CHOP、IRE1、Bax蛋白表达显著高于空白组(P<0.05),针刺组和西药组显著低于模型组(P<0.05);模型组胰腺CHOP、IRE1、Bax mRNA表达显著高于空白组(P<0.01),针刺组和西药组显著低于模型组(P<0.01);模型组Bcl-2 蛋白表达显著低于空白组(P<0.05),针刺组和西药组显著高于模型组(P<0.01),针刺组与西药组之间差异无统计学意义(P&gt;0.05);模型组Bcl-2 mRNA表达显著低于空白组(P<0.01),针刺组和西药组显著高于模型组(P<0.01),针刺组与西药组之间差异无统计学意义(P&gt;0.05)。结论:STZ糖尿病大鼠 胰腺组织存在内质网应激,IRE1-CHOP信号通路是抑制胰腺内质网应激的主要通路,针刺可以通过IRE1-CHOP信号通路改善胰腺组织内质网应激,抑制凋亡基因,保护胰腺组织。</p><p style="text-align: justify;">[关键词] 针刺 凋亡 糖尿病 需肌醇蛋白1</p><p><br/></p>
